They can replace Fusion Energy Disks that have already been placed and a disk can replace a Mystic Stone that has been placed already.

(For example, if the blue column was desired to have a Blue Stage 5 Fusion Energy Disc and the rest of the slots Mystic Stones, one would have to craft and apply all 5 blue discs, then replace the undesired stages with Mystic Stones.)
